Abstract:Protein metalation is a process through which metal compounds react with proteins and form adducts via coordination of metals to residue side chains. This process is at the basis of many biological events, including uptake, transport, storage, toxicity and mechanism of action of metal-based drugs. Knowledge of the protein metalation process at atomic level is important in the design and development of new metallodrug-based therapeutic approaches. In the last 15 years (1-7), using X-ray crystallography combined with other biophysical techniques, in my group we studied the protein metalation by Pt, Au, Ru, Rh, Ir, and V-based drugs. The main results of these studies will be summarized in this lecture. |
